A heads-up to English speakers: in Italian we say "La chiamata è di tua madre"; we use a different preposition.

Prepositions are one of the most difficult aspects to master in a language, as they is no 1:1 match between two languages.

This is one of the too many instances in which Duolingo translates literally, but the resulting sentence turns out wrong.

Does "la chiamata è da tua madre" instead of "la chiamata è di tua madre" has a meaning at all or is it simply not used?

The way I understand it:
la chiamata è da tua madre : the call is from your mother.
la chiamata è per tua madre : the call is for your mother. (e.g. an incoming call on a landline)
la chiamata è a tua madre : the call is to your mother.

As Sofia222677 indicated, the preposition required in this case is "di," not "da."
